I knew it immediately but thought surely someone else would answer this one. Since they haven't, I'll do the honors!
Season 5 finale, episode 24, Watershed.
Kate came to the park expecting that Castle was going to break up with her and instead, he proposed! We had to wait that whole summer to hear her answer in the premier of season 6.

This is SNEAKY! If it isn't Watershed, then it has to be Season 6, episode 1, Valkyrie, when Kate said, "Yes, I'll marry you." The only clue between the 2 scenes is Kate's big smile. When I went back and watched it again, in Watershed, she was all tense and there was no smile.
